目的测定八棱丝瓜蛋白2(LF-2)对小鼠黑色素瘤B16细胞的诱导分化作用。方法以MTT法测定LF-2对B16细胞的增殖抑制作用,以形态学、黑色素含量及细胞周期变化为指标,观察LF-2诱导B16细胞分化的作用。结果LF-2对B16细胞具有明显的增殖抑制作用,并使B16细胞停滞于G1期;细胞生长缓慢,平铺不重叠,甚至形成网状结构,呈正常上皮样细胞分化,细胞质内细胞器丰富,黑色素小体明显增多;黑色素含量显著增加。结论LF-2对B16细胞具有明显的诱导分化作用。
Objective To determine the induction and differentiation of Bailucaoguin 2 (LF-2) on mouse melanoma B16 cells. Methods The inhibitory effect of LF-2 on the proliferation of B16 cells was determined by MTT assay. The morphological, melanin content and cell cycle were used as indexes to observe the effect of LF-2 on differentiation of B16 cells. Results LF-2 could significantly inhibit the proliferation of B16 cells and arrest the B16 cells in G1 phase. The cells grew slowly and did not overlap and even formed reticular formation. The cells were normal epithelial-like cells with abundant organelles, Melanosomes increased significantly; melanin content increased significantly. Conclusion LF-2 has a significant induction and differentiation effect on B16 cells.
