目的研究梅连消渴胶囊对Ⅱ型糖尿病大鼠糖代谢的影响。方法高脂饲料喂养雄性SD大鼠5周后,腹腔注射小剂量链脲佐菌素(30 mg.kg-1),建立类似人类Ⅱ型糖尿病大鼠模型。动物随机分为模型对照组,盐酸二甲双胍组,梅连消渴胶囊高、中、低剂量组及空白对照组,连续灌胃给药4周。给药期间每周测定体重,每天记录动物的饮水量和摄食量;实验结束后测定空腹血糖水平(FBG)、口服糖耐量(OGTT)。末次给药后禁食12 h,腹主动脉采血测血清胰岛素(FINS)、糖化血红蛋白(HbAlc)。结果与模型组相比,梅连消渴胶囊高、中剂量组能显著降低糖尿病大鼠的FBG和FINS含量,同时改善动物摄食量、饮水量(P<0.01),降低HbAlc、胰岛抵抗指数(HOMA-IR)及OGTT曲线下面积(P<0.05),能明显提高胰岛敏感指数(ISI),高剂量组与二甲双胍组比较无显著性差异。结论梅连消渴胶囊对实验性Ⅱ型糖尿病大鼠糖代谢具有较好的调节作用,同时改善大鼠饮水及进食状况,降低大鼠胰岛素抵抗指数。
Objective To investigate the effect of Meiling Xiaoke Capsule on glucose metabolism in type 2 diabetic rats. Methods Male SD rats fed with high-fat diet for 5 weeks were injected intraperitoneally with low-dose streptozotocin (30 mg · kg-1) to establish a rat model of human type Ⅱ diabetes mellitus. The animals were randomly divided into model control group, metformin hydrochloride group, Meiling Xiaoke Capsule high, medium and low dose groups and blank control group, continuous gavage for 4 weeks. The body weight was measured weekly during the administration, and the water intake and food intake of animals were recorded every day. Fasting blood glucose (FBG) and oral glucose tolerance (OGTT) were measured after the experiment. After the last administration of fasting for 12 h, abdominal aorta blood test for serum insulin (FINS), glycosylated hemoglobin (HbAlc). Results Compared with the model group, Meiling Xiaoke Capsule high and middle dose groups could significantly reduce the FBG and FINS content of diabetic rats, and improve the food intake and water intake of animals (P <0.01), reduce the levels of HbA1c, HOMA-IR) and the area under the curve of OGTT (P <0.05), and significantly increased ISI. There was no significant difference between high dose group and metformin group. Conclusion Mei Lian Xiaoke Capsule has a good regulating effect on glucose metabolism in experimental type 2 diabetic rats, and at the same time it can improve drinking water and food intake of rats and decrease insulin resistance index in rats.
