目的:考察淫羊藿炮制前后的不同提取物对血管紧张素转化酶(ACE)、α-葡萄糖苷酶抑制活性及亚硝酸根清除活性的变化。方法:采用高效液相色谱法比较淫羊藿炮制前后的不同提取物对ACE的抑制作用,采用分光光度法测定其对亚硝酸盐的清除率以及对α-糖苷酶的抑制活性。结果:对ACE的抑制活性,只有石油醚提取物表现出了活性,并且未经炮制的淫羊藿明显强于炮制后的。对α-糖苷酶的抑制活性,未经炮制的淫羊藿二氯甲烷、乙酸乙酯、甲醇、水提取物及其水煎液均有活性;炮制后淫羊藿的二氯甲烷提取物则无活性,且其他3个提取物和水煎液的活性降低,但不明显。对亚硝酸根的清除活性,炮制后的淫羊藿石油醚、二氯甲烷、乙酸乙酯提取物和水煎液均表现一定的活性,未经炮制的淫羊藿的活性主要体现在二氯甲烷、乙酸乙酯、甲醇提取物和水煎液,其中水煎液比任何一个提取物的活性都好。本文也对炮制前后淫羊藿不同提取物、水煎液中淫羊藿苷的含量做了对比,结果显示淫羊藿苷可以在乙酸乙酯、甲醇、水提取物和水煎液中被检测到,炮制后淫羊藿的各个提取物的淫羊藿苷的含量均低于未炮制的。结论:淫羊藿经炮制后,其有效成分淫羊藿苷含量有所降低,且其对ACE、α-葡萄糖苷酶及亚硝酸根的抑制活性均有所降低。
Objective: To investigate the changes of angiotensin-converting enzyme (ACE), α-glucosidase inhibitory activity and nitrite scavenging activity of different extracts of epimedium before and after processing. Methods: HPLC was used to compare the inhibitory effect of different extracts of Epimedium before and after processing on the ACE inhibition. The removal rate of nitrite and the inhibitory activity of α-glucosidase were measured by spectrophotometry. Results: The ACE inhibitory activity, only the petroleum ether extract showed activity, and unprocessed Epimedium was significantly stronger than the processed. Inhibitory activity of α-glucosidase, unprocessed Epimedium dichloromethane, ethyl acetate, methanol, water extract and its decoction were active; Epimedium after processing the dichloromethane extract then No activity, and the other three extracts and decoction decreased activity, but not obvious. The removal activity of nitrite, processed epimedium petroleum ether, methylene chloride, ethyl acetate extract and decoction showed some activity, the unprocessed Epimedium activity mainly reflected in the dichloro Methane, ethyl acetate, methanol extract and decoction, decoction more than any one of the activity of the extract are good. CONCLUSION: Epimedium saponins has a reduced content of icariin after being processed, and its inhibitory activity on ACE, α-glucosidase and nitrite decreased.
