目的 探讨茶多酚对原代培养的皮肤角朊细胞和成纤维细胞生长作用的影响。方法 利用原代培养的两种主要的皮肤细胞———角朊细胞和成纤维细胞,采用丙酮酸比色法、TBA比色法和DTNB(双硫代对硝基苯甲酸)法测定细胞培养的上清液胞浆酶(LDH)释放情况、脂质过氧化产物(MDA)、谷胱甘肽过氧化物酶(GSHPx),并测定培养细胞的生长情况,对茶多酚在皮肤中可能起到的作用进行评价。结果 在加入茶多酚后不同时间,发现茶多酚具有保护细胞膜的功能,即减少LDH的释放、减少MDA产生、增加GSHPx含量、促进细胞生长。结论 体外实验表明茶多酚具有促进皮肤细胞生长、抗脂质过氧化的功效,其最低有效作用浓度为0.05%~0.1%。
Objective To investigate the effect of tea polyphenols on primary cultured keratinocytes and fibroblasts. Methods Using primary cultured keratinocytes and fibroblasts, pyruvate colorimetric method, TBA colorimetric method and DTNB (dithiobis p-nitrobenzoic acid) method were used to determine the cell culture (LDH), lipid peroxidation (MDA) and glutathione peroxidase (GSH-Px) in the supernatant were measured. Results After adding tea polyphenols at different times, it was found that tea polyphenols have the function of protecting the cell membrane, that is, reducing the release of LDH, decreasing the production of MDA, increasing the content of GSHPx, and promoting the cell growth. Conclusion In vitro experiments show that tea polyphenols can promote skin cell growth, anti-lipid peroxidation effect, the minimum effective concentration of 0.05% to 0.1%.
