目的 探讨不同方式骨髓移植的移植物抗宿主反应。方法 受体C57BL/ 6小鼠以60 Co辐射 ,经由尾静脉注入供体细胞。观察GVHD一般反应及病理学改变 ;通过解剖镜下计数脾结节、半固体琼脂法检测CFU -GM及常规计数外周WBC变化进行造血功能测定。G显带技术测定不同移植方式的嵌合体。结果 同基因与混合半相合组30d存活率高于其他两组 ,异基因组GVHD反应较强 ,半相合组次之 ,混合半相合组最弱 ,移植 13d时同基因组与混合半相合组脾结节无明显差别 ,但均高于异基因或半相合组 ,第 18天时混合半相合嵌合体与半相合组无明显差异 ,第 30天时混合半相合组嵌合体较高。异基因、半相合、混合半相合及同基因组的体内混合淋巴细胞反应程度 ,依次从强到弱。除异基因组的外周血WBC及骨髓CFU -GM较低外 ,其余各组无明显差别。结论 半相合与混合半相和骨髓移植可以减少GVHD反应
Objective To investigate the graft-versus-host reaction in different ways of bone marrow transplantation. Methods Recipient C57BL / 6 mice were irradiated with 60 Co and donor cells were injected via tail vein. The general response and pathological changes of GVHD were observed. The hematopoietic function of CFU-GM and routine peripheral WBC were measured by dissecting microscopic spleen nodules and semi-solid agar. G-banding technique to determine chimerism for different transplantation modalities. Results Compared with the other two groups, the survival rate of the same gene and mixed semi-phase group was higher than that of the other two groups. The response of GVHD in allogeneic group was stronger than that of the other groups. No significant difference was found, but all of them were higher than that of allogeneic or semi-phase group. On the 18th day, there was no significant difference between mixed half-phase chimera and half-phase chimerism. On the 30th day, the mixed half-phase chimeric chimera was higher. Heterogeneity, semi-coincidence, mixed half-occlusion and syngeneic mixed lymphocyte reaction in vivo, followed by strong to weak. In addition to different genomic peripheral blood WBC and bone marrow CFU-GMM lower, the rest of the group no significant difference. Conclusion Semi-phase and mixed phase and bone marrow transplantation can reduce the GVHD response
