目的:表面增强激光解吸离子化(surface enhanced laser desorption/ionization,SELDI)技术的重复性和稳定性进行分析。方法:将17名患者分成A、B两组,A组12名肿瘤患者清晨空腹血,离心后分成两份:一份立即行SELDI技术检测,另一份-80℃保存一周后再行检测;B组5名肿瘤患者连续两天各采血一次,并于采血当天行SELDI技术检测。所捕获的指纹经Biomarker Wizard3.1、ProteinChip3.2和Biomarker Pattern软件分析有无差异指纹。结果:两种采样方法都发现了差异指纹,其中有部分差异指纹在两种采样方法的检测中都出现,所有差异指纹M/Z(质荷比)由731到1167。结论:SELDI技术的确存在有重复性差、系统不稳定因素,但都发生在M/Z小分子量位段,对大位段数据分析未发现有明显影响。
Objective: To analyze the repeatability and stability of surface enhanced laser desorption / ionization (SELDI) technique. Methods: A total of 17 patients were divided into two groups: group A and group B. Twelve fasting blood samples from group A were divided into two groups: one was tested immediately by SELDI technique and the other was stored at -80 ℃ for one week. Five cancer patients in Group B took blood every two consecutive days and tested by SELDI technique on the day of blood sampling. The captured fingerprints were analyzed by Biomarker Wizard3.1, ProteinChip3.2 and Biomarker Pattern software for the presence or absence of differential fingerprints. RESULTS: Differential fingerprints were found in both sampling methods. Some of the differential fingerprints were detected in both sampling methods, and all the differential fingerprints M / Z (mass to charge ratio) ranged from 731 to 1167. Conclusion: The SELDI technique does have poor reproducibility and system instability, but all occur in the M / Z low molecular weight region. There is no obvious effect on the data from the large region.
