目的观察IL-17在日本血吸虫感染小鼠中对抗体及Th细胞免疫应答的影响。方法在小鼠日本血吸虫感染前后,连续多次腹腔注射rm-IL-17细胞因子(实验组1)或PBS(对照组1),连续多次腹腔注射anti-IL-17中和抗体(实验组2)或同型抗体(对照组2)。运用流式细胞术,采用表面分子、胞内因子同时染色的三色标记法,对日本血吸虫感染小鼠脾脏、淋巴结中的Th17、Th1和Th2细胞亚群占CD4+T细胞的比例进行观察;同时采用ELISA法检测各组小鼠血清中血吸虫抗原特异性总IgG及亚类IgG1和IgG2a的水平。结果实验组1 CD4+T细胞中的Th17、Th1和Th2细胞亚群的比例分别是对照组1的1.09倍(P(0.05)、1.17倍(P(0.05)和1.15倍(P(0.05);实验组2 CD4+T细胞中的Th17、Th1和Th2细胞亚群的比例分别是对照组2的1.01倍(P(0.05)、1.11倍(P(0.05)和0.98倍(P(0.05)。rm-IL-17注射后仅可溶性虫卵抗原(SEA)特异性IgG1水平显著下降(P(0.05)、可溶性成虫抗原(SWA)特异性IgG2a的水平升高(P(0.05),其他抗体水平均无显著变化;anti-IL-17中和抗体注射后,SEA特异性总IgG(P(0.05)、IgG2a(P(0.01)和SWA特异性IgG2a(P(0.01)水平显著升高,其他抗体水平均无显著变化。结论IL-17可能不会显著增强或降低宿主的血吸虫特异性Th细胞应答,而可能对IgG类抗体的产生具有某种程度的抑制作用。
Objective To observe the effect of IL-17 on the immune response of Th cells in mice infected with Schistosoma japonicum. Methods Before and after the infection of Schistosoma japonicum, anti-IL-17 neutralizing antibody was injected into the abdominal cavity continuously for several consecutive times by intraperitoneal injection of rm-IL-17 cytokine (experimental group 1) or PBS (control group 1) 2) or isotype antibody (control group 2). The ratio of Th17, Th1 and Th2 subsets in spleen and lymph node of CD4 (superscript +) T lymphocytes in CD4 (superscript +) T lymphocytes of mice infected with Schistosoma japonicum was observed by flow cytometry and trichrome labeling method using surface molecule and intracellular factor simultaneously. At the same time, the levels of total IgG and IgG1 and IgG2a in serum of schistosoma japonicum were detected by ELISA. Results The proportions of Th17, Th1 and Th2 subsets in CD4 + T cells of experimental group 1 were 1.09 times (P (0.05), 1.17 times (P <0.05) and 1.15 times higher than that of control group 1 respectively (P (0.05); The proportion of Th17, Th1 and Th2 subsets in CD4 + T cells in experimental group 2 was 1.01-fold (P (0.05), 1.11-fold (P> 0.05) and 0.98-fold After injection of IL-17, the levels of specific IgG1 in soluble egg antigen (SEA) were significantly decreased (P <0.05), while the levels of specific IgG2a in soluble adult antigen (SWA) (P (0.05), IgG2a (P (0.01)) and SWA-specific IgG2a (P (0.01)) were significantly increased after anti-IL-17 neutralizing antibody injection No significant change.Conclusion IL-17 may not significantly enhance or reduce the host-specific Schistosome-specific Th cell response, and may have some degree of inhibition of the production of IgG antibodies.
