目的:研究蒙药“连翘-4味汤散”对大鼠幽门结扎引起急性肝损伤的影响。方法:采用幽门结扎法制备大鼠急性肝损伤模型。造模16 h后分别检测正常组、模型组、蒙药连翘-4味汤散低、中、高剂量组(1.3,2.6,3.9 g·kg-1)ig给药,连续10 d后血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)、单胺氧化酶(MAO)、一氧化氮(NO)、一氧化氮合酶(NOS)、胆碱酯酶(CHE)、尿素氮(BUN)、总胆固醇(CHO)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)的水平以及肝指数和胃匀浆胃蛋白酶活性。结果:与正常组比较,模型组血清AST,ALT,BUN,NOS均明显增高(均P<0.01),CHE,NO明显降低(P<0.05),MAO,CHO,TG,LDL-C含量肝脏指数和胃蛋白酶活性均明显增高(P<0.05);与模型组比较,连翘-4味汤散中、高剂量组血清AST,ALT,NOS,MAO,CHO,TG,LDL-C水平及肝脏指数和胃蛋白酶活性均有明显降低(P<0.05,P<0.01),CHE,NO明显增高(分别为P<0.05),低剂量组对各种指标有好转趋势,但无显著差异。模型组呈肝细胞萎缩、重度脂肪变性、水肿;连翘-4味汤散中剂量组与正常组基本接近,高剂量组有少量脂肪变和轻度水肿,低剂量组与模型组无明显差异。结论:蒙成药连翘-4味汤散对幽门结扎引起的急性肝损伤具有明显的保护作用,且中剂量效果最好。
Objective: To study the effects of Mongolian medicine “Forsythia-Fortuner Decoction” on acute hepatic injury induced by pylorus ligation in rats. Methods: Acute hepatic injury model in rats was established by pylorus ligation. After 16 h of modeling, normal group, model group and Mongolian medicine Forsythia-Fortucaiandan Decoction were respectively administered with low, middle and high dosage of ig (1.3, 2.6, 3.9 g · kg-1) ALT and AST, MAO, NO, NOS, CHE, BUN, , Total cholesterol (CHO), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and liver index and gastric homopaus pepsin activity. Results: The levels of AST, ALT, BUN and NOS in model group were significantly higher than those in normal group (all P <0.01), while CHE and NO were significantly decreased (P <0.05) (P <0.05). Compared with the model group, the levels of AST, ALT, NOS, MAO, CHO, TG, LDL-C and the liver index (P <0.05, P <0.01). CHE and NO were significantly increased (P <0.05, respectively). The low dose group showed a trend of improvement for all indexes, but there was no significant difference between the two groups. The model group showed hepatocyte atrophy, severe steatosis and edema; the forsythia-4 and Weiweisan medium dose group was close to the normal group, while the high dose group had a little steatosis and mild edema. There was no significant difference between the low dose group and the model group . Conclusion: Mongolian medicine Forsythia-4 Wentang San has obvious protective effect against acute hepatic injury caused by pylorus ligation, and the middle dose is the best.
