目的 研究不同类型新月体肾炎患者肾小球中C-C类趋化因子单核细胞趋化蛋白-1(MCP-1)、巨噬细胞炎性蛋白-1α和β(MIP-1α、MIP-1β)的表达,分析其与肾小球中浸润的单核细胞及Bowman 囊壁是否断裂之间的关系,从而探讨其在新月体肾炎发生和发展中的作用。方法 选择不同类型新月体肾炎患者 32例,其中 Ⅰ型 8例,Ⅱ型 12例,Ⅲ型 12例。应用免疫组织化学方法分别观察细胞性和纤维细胞性新月体肾炎的肾小球中 CD68、MCP-1、MIP-1α和 MIP-1β的表达。结果 3种类型新月体肾炎肾小球中 CD68、MCP-1、MIP-1α和 MIP-1β的表达均明显增加;新月体肾炎肾小球中MCP-1和MIP-1α的表达与单核细胞浸润之间呈正相关(相关系数r分别为0.568和0.749,P均<0.01);Ⅰ型新月体肾炎不仅肾小球中MCP-1和MIP-1α的表达均高于Ⅱ型和Ⅲ型新月体肾炎,而且Bowman囊壁断裂的发生率也较Ⅱ型和Ⅲ型新月体肾炎患者明显升高(P<0.05)。结论 肾小球内MCP-1、MIP-1α和MIP-1β表达增加是新月体形成的重要始动因素,MCP-1和MIP-1α不仅介导新月体肾炎肾小球中单核细胞的?
Objective To investigate the expression of C-C chemokine monocyte chemoattractant protein-1 (MCP-1), macrophage inflammatory protein-1α and β (MIP-1α, MIP) in glomerulus of different types of patients with crescentic glomerulus 1β) expression in glomerular infiltration of mononuclear cells and Bowman’s wall is the relationship between the fracture, to explore its role in the occurrence and development of crescentic glomerulonephritis. Methods 32 cases of different types of crescentic glomerulonephritis were selected, including 8 cases of type Ⅰ, 12 cases of type Ⅱ and 12 cases of type Ⅲ. Immunohistochemistry was used to observe the expression of CD68, MCP-1, MIP-1α and MIP-1β in glomerulus of cellular and fibrocystic crescentic nephritis. Results The expressions of CD68, MCP-1, MIP-1α and MIP-1β in the glomerulus of three types of crescentic glomeruli were significantly increased. The expressions of MCP-1 and MIP-1α There was a positive correlation between the expression of MCP-1 and MIP-1α in glomerulus (r = 0.568 and 0.749, P <0.01, respectively) Higher than those of type II and III crescentic glomerulonephritis, and the incidence of Bowman’s wall fracture was also significantly higher than those of type II and III crescentic glomerulonephritis (P <0.05).
