目的研究淋巴瘤患者三氧化二砷治疗前后外周血单核细胞端粒酶逆转录酶表达的变化以及可能的机制。方法用实时荧光定量PCR方法检测22例患者,比较hTERT的相对值NhTERT(NhTERT=100×hTERT拷贝数/GAPDH拷贝数)的差异,并与正常组20例进行比较。结果三氧化二砷治疗前后淋巴瘤外周血单核细胞hTERT基因表达均高于正常组,治疗后外周血hTERT基因表达有一定的下降。结论淋巴瘤患者外周血单核细胞hTERT基因表达对于诊断有一定的意义,诱发hTERT基因表达下降,降低端粒酶活性可能是As_2O_3治疗淋巴瘤的机制之一。
Objective To study the changes of telomerase reverse transcriptase expression in peripheral blood mononuclear cells before and after treatment with arsenic trioxide in patients with lymphoma and its possible mechanism. Methods Twenty-two patients were detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR). The relative value of hTERT was compared with that of NhTERT (NhTERT = 100 × hTERT copy number / GAPDH copy number). Results The expression of hTERT gene in peripheral blood mononuclear cells before and after treatment with arsenic trioxide was higher than that in normal group. The expression of hTERT gene in peripheral blood decreased after treatment. Conclusion The expression of hTERT gene in peripheral blood mononuclear cells of patients with lymphoma is of certain significance for the diagnosis. The decrease of hTERT gene expression and the decrease of telomerase activity may be one of the mechanisms of As 2 O 3 in the treatment of lymphoma.
