目的:建立RP-HPLC法测定乐脉颗粒中丹参素、原儿茶醛、芍药苷和阿魏酸4种有效成分的含量。方法:采用Dia-monsil-C18(200mm×4.6mm,5μm)色谱柱,以乙腈-0.1%磷酸为流动相梯度洗脱,流速1.0mL·min-1,柱温30℃,检测波长230nm。结果:丹参素、原儿茶醛、芍药苷和阿魏酸浓度分别在22.0~440,1.90~38.0,71.0~1.42×103,0.640~12.8μg·mL-1范围内与峰面积线性关系良好(r>0.9995);方法平均回收率(n=9)分别为100.8%,98.6%,101.1%,99.7%。结论:本方法简便、准确,重复性好,可用于同时测定乐脉颗粒中丹参素、原儿茶醛、芍药苷和阿魏酸的含量。
Objective: To establish a RP-HPLC method for the determination of four active components of Danshensu, protocatechuic aldehyde, paeoniflorin and ferulic acid in Lemai Granules. Methods: A Dia-monsil-C18 (200mm×4.6mm, 5μm) column was used. The mobile phase was acetonitrile-0.1% phosphoric acid. The flow rate was 1.0mL·min-1, the column temperature was 30°C, and the detection wavelength was 230nm. Results: The concentration of danshensu, protocatechuic aldehyde, peoniflorin and ferulic acid in the range of 22.0~440, 1.90~38.0, 71.0~1.42×103 and 0.640~12.8μg·mL-1 were linear with the peak area. (r>0.9995); The average recovery of the method (n=9) was 100.8%, 98.6%, 101.1%, and 99.7%, respectively. Conclusion: The method is simple, accurate and reproducible. It can be used to determine the content of Danshensu, protocatechuic aldehyde, paeoniflorin and ferulic acid in Lemai Granules.
