我们以肺螨病患者痰检时常见的混合螨种制作螨组织抗原片,现将其制作过程简要报道如下。 1 混合螨种的培养和纯化 从面粉厂收集地脚粉,经检查内有螨卵、幼虫和活的雌雄成螨,置25℃、相对湿度80％条件下培养;经鉴定所含螨种以粗脚粉螨、腐食酪螨、粉尘螨、椭圆食粉螨、纳氏皱皮螨和食虫狭螨为主,与肺螨病患者痰检的常见螨种相符合。培养至对数生长期后,将面粉置70目铜筛内径70目和120目两层迭加的铜筛过滤,其中间部分即120目筛内的部分置85目铜筛内,上方约25cm处以25w白炽灯照射,下方置盛有少许蒸馏水或0.65％盐水的平底容器,经2h待螨因受热爬出入水。以饱和盐水漂浮法分离,0.65％盐水离心清洗3～4次。 2 螨组织切片
