目的:阐述关节软骨组织工程修复的种子细胞及其应用的研究现状,对其来源、体外扩增、以及基因修饰进行了探讨,并提出其进一步应用的前景。资料来源:检索PubMed1994-01/2006-04与关节软骨组织工程修复的种子细胞相关的文章,检索词为“cartilage,joint,tissue engineering,stem cell,cell culture,gene modification”,并限定语言种类为英语。其他文章经清华同方和维普数据库检索具体的杂志名称和文章题名收集。资料选择:对收集到的资料进行初筛,选择与关节软骨组织工程修复的种子细胞相关的文章,筛除明显不符合原文的文章,对剩余的文献手工检索查找全文。资料提炼:共收集相关文章89篇,纳入34篇,排除55篇。资料综合:用于构建人工软骨组织的细胞主要有自体或异体软骨细胞、骨膜和软骨膜细胞、间充质干细胞以及胚胎干细胞,其均可于体外经特定的培养体系进行有效的培养和扩增,在一定条件下还可以经不同的策略和方法进行基因修饰,从而修复关节软骨缺损。结论:种子细胞以其自身多方面的特点在组织工程技术修复关节软骨缺损中发挥重要作用。
OBJECTIVE: To describe the status quo of seed cells repaired by articular cartilage tissue engineering and its application. The origin, in vitro expansion and gene modification of the seed cells were discussed. The prospects for their further application were also discussed. Source: PubMed searched from January 1994 to April 2006 with articles related to articular cartilage tissue engineered seed cells. The search term was “cartilage, joint, tissue engineering, stem cell, cell culture, gene modification” English. Other articles by Tsinghua Tongfang and VIP database search magazine name and article title collection. Data selection: screening the collected data, selecting articles related to seed cells repaired by articular cartilage tissue engineering, screening out articles that obviously do not meet the original text, and manually searching the remaining articles for the full text. Data extraction: A total of 89 related articles were collected, including 34 articles and 55 articles. DATA SYNTHESIS: The cells used for constructing artificial cartilage tissue mainly include autologous or allogeneic chondrocytes, periosteum and perichondrocytes, mesenchymal stem cells and embryonic stem cells, which can be effectively cultured and expanded in a specific culture system in vitro , Under certain conditions can also be modified by different strategies and methods to repair the articular cartilage defects. Conclusion: The seed cells play an important role in the repair of articular cartilage defects by tissue engineering due to their own characteristics.
