目的观察刺山柑总生物碱对系统性硬皮病(SSc)小鼠结缔组织生长因子(CTGF)、转录因子Smad3及磷酸化细胞外信号调节激酶(pERK)水平的影响。方法 BALB/c小鼠背部皮下注射盐酸博莱霉素诱导SSc模型,给予225,450,900mg·kg~(-1)刺山柑总生物碱治疗后,ELISA法检测小鼠皮肤组织CTGF、Smad3和pERK水平。结果 450,900 mg·kg~(-1)刺山柑总生物碱可显著降低SSc小鼠皮肤组织CTGF水平(P<0.05或P<0.01),900 mg·kg-1可降低Smad3水平(P<0.01),对p ERK没有明显影响(P>0.05)。结论刺山柑总生物碱可调节SSc CTGF和Smad3的异常升高,改善SSc组织纤维化。
Objective To investigate the effects of total alkaloids from capers on the levels of connective tissue growth factor (CTGF), transcription factor Smad3 and phosphorylated extracellular signal-regulated kinase (pERK) in mice with systemic scleroderma (SSc). Methods BALB / c mice were subcutaneously injected with bleomycin hydrochloride to induce SSc model. The total alkaloids of capers of capparis were treated with 225,450 and 900 mg · kg -1. The levels of CTGF, Smad3 and pERK in skin of mice were detected by ELISA. . Results Total alkaloids at 450 and 900 mg · kg -1 could significantly reduce CTGF level in skin of SSc mice (P <0.05 or P <0.01), and decrease the level of Smad3 at 900 mg · kg -1 (P <0.01) ), No significant effect on p ERK (P> 0.05). Conclusion The total alkaloids of capers lead to the abnormal increase of SSc CTGF and Smad3 and the improvement of SSc tissue fibrosis.
