目的:研究复方SZ滴眼液(Compound shui zhi eye drop,SZ)和阿魏酸钠(Sodium ferulate,SF)对紫外线(Ultraviolet,UV)损伤的人晶状体上皮细胞抗氧化酶及其基因表达的影响。方法:采用UV照射体外培养的人晶状体上皮细胞(Human lens epithelial cell,HLEC),同时用SZ、SF和吡喏克辛钠滴眼液(Pirenoxine sodium,PS)与HLEC共同孵育,并检测HLEC中超氧化物歧化酶(Superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(Glutathi-one peroxidase,GSH-px)和过氧化氢酶(Catalase,CAT)的活性和SODmRNA、GSH-pxmRNA和CATmRNA的基因表达。结果:UV组的HLEC抗氧化酶活性和基因表达均比对照组显著降低(P<0.01)。SZ、SF和PS组的SOD、GSH-px和CAT的活性显著高于UV组(P<0.01,P<0.05);SODmRNA、GSH-pxmRNA和CATmRNA的表达也显著高于UV组(P<0.01)。结论:SZ、SF和PS均能防护UV照射引起的HLEC氧化损伤,SZ和SF的作用优于PS。
OBJECTIVE: To study the effects of compound SJ and sodium ferulate on antioxidant enzymes and their gene expression in human lens epithelial cells damaged by ultraviolet (UV) . Methods: Human lens epithelial cells (HLECs) cultured in vitro were exposed to UV light. SZ, SF and Pirenoxine sodium (PS) were incubated with HLEC and HLEC The activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-px) and catalase (CAT) and the activities of SODmRNA, GSH-pxmRNA and CAT mRNA gene expression. Results: Compared with the control group, the anti-oxidative enzyme activity and gene expression of HLEC in UV group were significantly lower (P <0.01). The activities of SOD, GSH-px and CAT in SZ, SF and PS groups were significantly higher than those in UV group (P <0.01, P <0.05). The expressions of SODmRNA, GSH-pxmRNA and CAT mRNA were also significantly higher than those in UV group ). Conclusion: SZ, SF and PS can protect the oxidative damage of HLEC induced by UV irradiation. The effect of SZ and SF is better than that of PS.
