目的:建立测定胆宁片中金丝桃苷含量的高效液相色谱方法。方法:应用高效液相色谱法,采用Venusil XBP-C18色谱柱(4.6mm×250mm,5μm);流动相为乙腈-0.05%甲酸(三乙胺调pH至5.0),梯度洗脱;柱温为25℃,检测波长:358nm,流速:0.8mL.min-1,进样20μL。结果:金丝桃苷进样浓度在13.26-132.62μg.mL-1范围内呈现良好的线性关系,回归方程:Y=17 229X-9 983,r=0.999 6,加样平均回收率为98.13%,RSD为1.41%。结论:本法操作简便,结果准确稳定,可为胆宁片的质量控制方法提供依据。
Objective: To establish a high performance liquid chromatographic method for the determination of hyperin content in Danning tablets. Methods: High performance liquid chromatography (HPLC) was performed on a Venusil XBP-C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of acetonitrile-0.05% formic acid 25 ℃, detection wavelength: 358nm, flow rate: 0.8mL.min-1, injection 20μL. Results: The concentration of hyperin in the range of 13.26-132.62μg.mL-1 showed a good linear relationship, the regression equation: Y = 17 229X-9 983, r = 0.999 6, the average recovery rate was 98.13% , RSD is 1.41%. Conclusion: The method is simple, accurate and stable, which can provide the basis for the quality control method of Danning tablets.
