酒精性肝损伤已成为近年高发疾病之一,如何预防过度饮酒导致的肝脏疾病已成为人们关注的热点.花青素可预防酒精性肝损伤,但作用靶点仍不清楚.本实验通过建立慢性酒精性肝损伤动物模型,系统探讨了紫甘薯和紫土豆中的花青素对小鼠慢性酒精性肝损伤的作用机制,通过检测酒精性肝损伤的标记物:天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、肝脏总胆固醇(TCH)、甘油三酯(TG)、丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽还原酶(GSH)的含量,确定花青素的保护作用,通过检测肿瘤坏死因子(TNF-α)、7型干扰素(IFN-γ)、血管细胞粘附因子-1 (VCAM-1)、趋化因子(CXCL-1)、Toll样受体-4(TLR-4)、信号传导与激活因子(STAT-3)和核转录因子(NF-κB) mRNA的表达含量及细胞色素P450 2El酶(CYP2E1)蛋白的表达含量,探索其作用靶因子.
