Major isoflavonoids of Astragali Radix (AR) arc calycosin, formononctin, L-3-hydroxy-9,10-dimethoxypterocarpan, (3R)-7,2-dihydroxy-3,4-dimethoxyisoflavanand their glycosides.The health benefits of these isoflavonoids at in vitro study have been reported, however, the results of direct analysis of isoflavonoids and their metabolites in biofluids after consumption of AR-containing decoction are scarce.Purpose: The aim of this study was to develop a sensitive HPLC-MS/MS method to directly screen and analyze the isoflavonoids and their conjugated metabolites in rabbit plasma after orally administered A.mongholicus extract.Method: The HPLC-MS system consisted of a Waters Alliance 2690 apparatus (Milford, MA, USA) and a ThermoQuest Finnigan TSQ (Triple-Stage Quadruple) mass spectrometer (San Jose, CA, USA) equipped with a Finnigan ESI interface.
