I will discuss two methods we use to produce human monoclonal antibodies and cognate antigens for cancer irnmunotherapy.1)Identify a cell surface receptor expressed selectively on the endothelial cells of tumor blood vessels and/or tumor cells, andthe ligand for the receptor.Synthesize an antibody chimera composed of the ligand conjugated to the human IgG1-Fc for passive irnmunotherapy.2) Construct a phage-display library expressing VH domains cloned from peripheral blood lymphocytes and lymph nodes of cancer patients.Screen the libraries for phage that bind selectively to human tumor lines corresponding to the patients type of tumor.Synthesize the selected VH-Fc antibodies for passive immunotherapy.Clone the tumor antigen recognized by the antibody for use as a vaccine.
