目的:建立测定人血清中头孢拉定浓度的高效液相色谱法(HPLC)。方法:血清经高氯酸处理后进样,采用Bio-Rad-C18不锈钢柱,以甲醇一醋酸盐缓冲液(25∶75,v/v)为流动相,茶碱为内标,在波长254nm处测定。结果:头孢拉定的线性范围为5~100mg/L,平均回收率为(1003±23)%,日内、日间精密度RSD均小于3%。结论:本法简便、可靠、适用于临床监测。
Objective: To establish a high performance liquid chromatography (HPLC) for the determination of cefradine in human serum. Methods: Serum was injected with perchloric acid solution. Bio-Rad-C18 stainless steel column was used with methanol-acetate buffer (25:75, v / v) as mobile phase and theophylline as internal standard. 254nm at the determination. Results: The linear range of cefradine was 5 ~ 100 mg / L with the average recovery of (1003 ± 23)%. The intra-day and inter-day RSD were less than 3%. Conclusion: This method is simple, reliable and suitable for clinical monitoring.
