Top-down and bottom-up strategies are widely adopted in proteome study.However, no matter by which means, the enzymatic digestion is indispensable so that proteins could be identified by MS.Up till now, free solution digestion is commonly used for protein digestion, which suffers disadvantages such as rather long incubation time, high liability for enzyme autodigestion and disability to achieve on-line hyphenation with the separation and identification systems.Therefore, it is requisite to develop novel enzymatic reactors.In our recent work, various microenzymatic reactors were prepared by immobilizing trypsin on different substrates, such as packing materials and monoliths, which showed great potential in proteome study.
