[目的]小菜蛾Plutella xylostella (L.)是十字花科蔬菜的重要害虫,目前对包括氯虫苯甲酰胺在内的多种杀虫剂都已产生了高水平的抗性.现有研究已经表明多个蛋白编码基因参与调控了小菜蛾对氯虫苯甲酰胺的抗药性,但有关氯虫苯甲酰胺抗药性转录后调控机制的报道还相对较少.本研究系统鉴定了与小菜蛾对氯虫苯甲酰胺抗药性相关的microRNA (miRNA).[方法]构建了一个小菜蛾敏感品系(CHS)以及两个氯虫苯甲酰胺抗性品系(CHR,ZZ)的miRNA文库,并进行高通量测序分析,运用生物信息学软件对miRNA及其靶基因进行预测.[结果]通过测序共鉴定得到了481个已知的小菜蛾miRNAs和30个新的miRNAs.在CHR和CHS之间共检测到31个差异表达的miRNAs,在ZZ和CHS之间检测到176个差异表达的miRNAs,其中有21个差异表达的miRNA是两个抗性品系共有的.利用miRNA靶标预测软件miRanda和RNAhybrid,在186个差异表达的miRNA中共预测到了2802个靶标基因,其中包含了多条解毒酶基因.进一步应用qRT-PCR对部分差异表达的miRNA及其靶标基因的表达情况进行了验证.[结论]本研究首次系统的鉴定了小菜蛾氯虫苯甲酰胺抗性相关的miRNA,为进一步在小菜蛾和其他目标昆虫中研究miRNA介导的氯虫苯甲酰胺抗性机制提供了重要线索.
