目的 探讨在肝卵圆细胞自我更新及增殖的调控中Bmi1是否通过Ink4a/Arf通路发挥作用.方法 利用前期构建的野生型小鼠、Bmi1肝脏特异性敲除(Bmi1alb-/-)小鼠以及Bmi1、Ink4a/Arf双基因肝脏特异敲除(Bmi1alb-/-；Ink4a/Arfalb-/-)小鼠,用PCR技术及Western印迹法对敲除效率做了验证(90％).通过给3种品系小鼠连续喂食3周0.1％DDC食物来诱导小鼠肝卵圆细胞扩增模型.利用免疫组织化学技术特异性地给小鼠肝卵圆细胞染色(肝脏卵圆细胞特异性标示物Epcam),并通过Epcam的表达情况来分析不同品系小鼠肝脏中卵圆细胞的扩增情况.结果 与没有喂食0.1％DDC食物的野生型小鼠相比,喂食3周0.1％DDC食物的野生型小鼠肝脏中卵圆细胞数目明显增加(P＜0.05).在喂食3周0.1％DDC食物后,Bmi1alb-/-小鼠肝脏中肝卵圆细胞数明显少于野生型小鼠肝脏中卵圆细胞数(P＜0.05)；而Bmilalb-/-；Ink4a/Arfalb-/-小鼠肝脏中卵圆细胞数多于Bmi1alb-/-小鼠肝脏中卵圆细胞数(P＜0.05),但少于野生型小鼠肝脏中卵圆细胞数(P＜0.05).结论 Bmil促进肝脏卵圆细胞的自我更新及增殖是部分依赖Ink4a/Arf通路的,但除此之外还有其他通路有待发现.
