目的 探讨人玻璃体液对培养的人视网膜色素上皮 (retinal pigment epithelium,RPE)细胞和血管内皮细胞增生的影响。 方法 将人 RPE细胞和血管内皮细胞株 (vascular endothelial cell lines30 4 ,VEC30 4 )分别培养在 1∶ 8、1∶ 4、1∶ 2 (玻璃体液在总培养液中的体积比 )的人玻璃体条件培养液(vitreous- conditioned medium,VCM)中 ,其中 VCM分为有无血清 2组 ,在不同作用时间 (2~ 6 d) ,采用四唑盐 (tetrazolium,MTT)比色法检测 VCM对人 RPE细胞和 VEC30 4增生的影响。 结果 在有血清时 ,1∶ 4、1∶ 2的 VCM在培养的各时间段对 RPE细胞的促增生作用与对照组比较 ,差异有非常显著性意义(P<0 .0 1) ,对 VEC30 4的抑增生作用与对照组比较 ,差异有显著性意义 (P<0 .0 5 ) ;在无血清时 ,1∶ 2的VCM在培养 2 d时对 RPE细胞的促增生作用与对照组比较 ,差异有显著性意义 (P<0 .0 5 ) ,在培养 4、6 d时的差异有非常显著性意义 (P<0 .0 1)。 结论 一定体积分数的人 VCM促进人 RPE细胞的增生 ,抑制VEC30 4的增生 ,提示玻璃体液在增生性玻璃体视网膜病变 (proliferative vitreoretinopathy,PVR)和眼内血管增生性疾病中可能发挥不同的调控作用。
Objective To investigate the effects of human vitreous humor on the proliferation of cultured human retinal pigment epithelium (RPE) cells and vascular endothelial cells. METHODS: Human vitreous cells were cultured in a ratio of 1: 8, 1: 4 and 1: 2 (volume ratio of vitreous fluid to total medium) of human RPE cells and vascular endothelial cell lines 304 (VEC304) (VCM) were divided into two groups: serum-free group (VCM) and serum-free group (VCM) at different time points Effect of cells and VEC30 4 hyperplasia. Results In the presence of serum, VCM at a ratio of 1: 4 and 1: 2 promoted the proliferation of RPE cells at different time points in culture compared with the control group, and the difference was statistically significant (P <0.01). VEC30 4 compared with the control group, the difference was significant (P <0. 05); in serum-free, 1: 2 VCM cultured 2 days on RPE cells in the role of proliferation compared with the control group , The difference was significant (P <0. 05), in the culture of 4,6 d the difference was significant (P <0.01). Conclusion A certain volume fraction of human VCM promotes proliferation of human RPE cells and inhibits the proliferation of VEC30 4, suggesting that vitreous fluid may play different regulatory roles in proliferative vitreoretinopathy (PVR) and ocular vascular proliferative diseases.
