为探究苹果属富含多糖多酚物质的平邑甜茶总RNA提取的最优方法,本研究以平邑甜茶的幼嫩叶片、盛花期子房、花瓣和萼片为材料,利用常规CTAB法和试剂盒法(TIANGEN生化科技有限公司提供的RNAprep Pure多糖多酚植物总RNA提取试剂盒)分别提取平邑甜茶不同器官、组织的总RNA,并对RNA浓度和和电泳图谱进行比较分析。研究结果表明:利用天根生化科技公司提供的多糖多酚植物总RNA提取试剂盒可提取到纯度较高,完整性较好的总RNA,可以用于获取cDNA全长、RT-PCR及荧光定量RT-PCR等分子生物学实验。
In order to explore the optimal method for the extraction of total RNA from Malus hupehensis which is rich in polysaccharides and polyphenols, this study took the young leaves, full-flowering ovary, petals and sepals of Malus hupehensis as material, using conventional CTAB method and reagent The total RNA of different organs and tissues of P. euphorbiae was extracted by the box method (RNAprep Pure Polysaccharide and Polyphenol Total RNA Extraction Kit provided by TIANGEN Biochemical Technology Co., Ltd.). The RNA concentration and electrophoresis pattern were compared. The results showed that the total RNA extracted by Polysaccharides and Polyphenols Total RNA Extraction Kit from Tiangen Biochemical Technology Company can be used to extract total RNA with high purity and integrity and can be used to obtain full length cDNA, RT-PCR and fluorescence quantitative RT-PCR and other molecular biology experiments.
