目的观察益气通络丹 (YQTLD)含药血清对培养SD大鼠胸主动脉平滑肌细胞 (VSMC)的影响 ,探讨该药抗冠心病、脑梗塞等心脑血管疾病的作用机制。方法血清药理学方法制备含药血清 ;MTT法测定含药血清对VSMC增殖活性的影响 ;NO试剂盒测定含药血清对VSMC作用与NO的关系。结果YQTLD含药血清能显著降低培养的SD大鼠VSMCMTT法的D(()值 ,与空白血清组有显著性差异 (P <0 0 1) ;YQTLD含药血清组培养基中NO含量明显增多 ,与空白血清组有显著性差异 (P <0 0 1)。结论YQTLD有抗VSMC增殖的作用 ,此作用可能与NO有关 ,YQTLD可能通过抗VSMC增殖 ,促进NO释放来延缓AS的发生、发展 ,从而在冠心病、脑梗死等心脑血管疾病中发挥作用。
Objective To observe the effect of YQTLD-containing serum on cultured rat thoracic aorta smooth muscle cells (VSMC) and to explore the mechanism of this drug against cardiovascular and cerebrovascular diseases such as coronary heart disease and cerebral infarction. Methods Serum-pharmacological methods were used to prepare drug-containing serum; MTT assay was used to determine the effect of drug-containing serum on proliferation of VSMC; NO kit was used to determine the relationship between drug-containing serum and VSMC. Results YQTLD drug-containing serum could significantly reduce the D(() value) of MTT assay in cultured SD rat VSMCs, and there was a significant difference with the blank sera group (P < 0.01); YQTLD-containing serum increased the NO content in culture medium. There was a significant difference between the blank sera and the blank sera (P <0 01).Conclusion YQTLD has anti-VSMC proliferation effect, which may be related to NO. YQTLD may delay the proliferation of the VSMC and promote the release of NO to delay the occurrence and development of AS. Therefore, it plays a role in cardiovascular and cerebrovascular diseases such as coronary heart disease and cerebral infarction.
