目的:研究桃红消癓散对子宫肌瘤模型大鼠雌激素受体(ER)、孕激素受体(PR)及增殖细胞核抗原(PCNA)表达的影响。方法:采用雌孕激素负荷法建立大鼠子宫肌瘤动物模型,通过放免法检测血清雌孕激素水平,观察子宫组织病理改变,免疫组化法观察大鼠子宫平滑肌细胞ER,PR及PCNA的表达。结果:模型大鼠较正常大鼠子宫明显增大,血清性激素水平升高,子宫平滑肌细胞ER,PR及PCNA的表达明显高于正常大鼠。与模型对照组比较,桃红消癓散可降低血清性激素水平,改善子宫组织结构,抑制平滑肌细胞ER,PR及PCNA的表达。结论:桃红消癓散有治疗子宫肌瘤的作用,其机制可能与调节血中雌孕激素水平,抑制子宫平滑肌细胞ER,PR及PCNA的表达有关。
Objective: To investigate the effect of Taohong Xiaoyaosan on the expression of estrogen receptor (ER), progesterone receptor (PR) and proliferating cell nuclear antigen (PCNA) in uterine leiomyoma model rats. Methods: The animal model of rat uterine fibroids was established by estrogen-progestin-loaded method. The level of estrogen and progesterone in serum was detected by radioimmunoassay. The pathological changes of uterus were observed. The expression of ER, PR and PCNA in rat uterine smooth muscle cells were observed by immunohistochemistry . Results: Compared with normal rats, the uteri of model rats were significantly increased and the level of serum sex hormone was increased. The expression of ER, PR and PCNA in uterine smooth muscle cells was significantly higher than that in normal rats. Compared with the model control group, Taohong Xiaoyaosan can reduce serum sex hormone levels, improve uterine tissue structure, inhibit the expression of ER, PR and PCNA in smooth muscle cells. Conclusion: Taohong Xiaoyaosan has the therapeutic effect on uterine fibroids. The mechanism may be related to the regulation of estrogen and progesterone levels in blood and the inhibition of the expression of ER, PR and PCNA in uterine smooth muscle cells.
