目的研究不同浓度的山楂叶总黄酮对鹌鹑血脂和脂肪肝形成的防治作用。方法将60只雌、雄各半的鹌鹑分成6组:正常组(N)喂以普通饲料;高脂饮食模型组(M)喂以高胆固醇脂饲料,诱发形成脂肪肝模型;脂必妥药物对照组(ZBT)、山楂叶总黄酮低、中、高浓度实验组(HLF1、HLF2、HLF3)在喂高脂饲料的同时分别喂1g/kg体重ZBT、20mg/kg、40mg/kg、80mg/kg体重山楂叶总黄酮溶液各1mL。实验6个月后取动物血清、肝脏,检测动物血清、肝脏组织中总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷丙转氨酶(AST)、谷草转氨酶(ALT)的含量,制备鹌鹑肝脏组织石蜡切片和电镜切片观察其病理学变化。结果山楂叶总黄酮实验各组与模型组相比,其血清和肝脏匀浆液的TC、TG、LDL-C、MDA、AST、ALT等含量均显著下降(P<0.001),而HDL-C、GSH、SOD含量则明显上升(P<0.001),且山楂叶总黄酮的浓度呈明显的量效关系。石蜡切片和电镜切片也显示山楂叶总黄酮实验组动物肝脏中的脂质积累和脂肪肝病理学变化明显轻于模型组。结论山楂叶总黄酮具有明显降低血脂、减轻动物肝脏内各类脂质沉积的作用,还具有很好的抗氧化作用,能显著地保护肝脏组织的生理生化功能,对由高脂饮食引起的高血脂和脂肪肝具有明显的防治作用。
