目的 利用毛细管电泳 (CapillaryElectrophoresis ,CE)分离效果好、稳定性强的特点。建立毛细管电泳测定血清中唾液酸 (SialicAcid ,SA)的方法 (CE SA法 ) ,并与经典的化学比色法 (CH SA法 )进行对比研究 ,以达到快速、准确测定血清SA的目的。方法 在 2ml试管中分别加入 0 0 5mol/L硫酸 1ml和 10 0 μl血清 ,充分混合 ,然后试管紧塞 ,于 80℃水浴箱中水浴 30min ,试管冷却后离心 5min ,吸 1ml上清液于自动进样管中上机进样。结果 CE SA法检测范围为 1 2 0~2 80mmol/L ,线性方程为y =-0 .6 48± 6 .394x ,相关系数 0 .9993 ,平均回收率是 10 1.2 3 %。 113名交警唾液酸 (CE SA法 )均值为 (1.76± 0 .2 0 )mmol/L[(5 49.30± 3 .73)mg/L]。CH SA法测定均值为 (1.75± 0 .2 4)mmol/L ,两方法无显著性差异 (P>0 .0 5 )。结论 毛细管电泳能提供一种很好的分离、检测唾液酸的方法 ,值得推广
Objective To use Capillary Electrophoresis (CE) for separation and good stability. A method for the determination of sialic acid (SA) in serum by capillary electrophoresis (CESA method) was established and compared with the classical chemical colorimetric method (CHSA method) to achieve rapid and accurate determination of serum SA. Methods 0 ml of 5ml/L sulfuric acid 1ml and 100μl of serum were respectively added into 2ml test tubes, mixed thoroughly, and then the test tubes were tightly closed. The water bath was placed in a water bath at 80°C for 30min, the tubes were cooled and centrifuged for 5min, and 1ml of supernatant was sucked into the autosampler. Inject the sample into the tube. Results The detection range of CE SA method was 120 to 2 80 mmol/L, the linear equation was y = -0.66 ± 6 .394x, the correlation coefficient was 0.993, and the average recovery rate was 10 1.2 3%. The average value of the 113 traffic police sialic acids (CESA method) was (1.76 ± 0.20) mmol/L [(5 49.30 ± 3.73) mg/L]. The average value of CHSA assay was (1.75±0.24 mmol/L). There was no significant difference between the two methods (P>0.05). Conclusion Capillary electrophoresis can provide a good method for the separation and detection of sialic acid. It is worth promoting.
