目的:建立反相高效液相色谱法测定柴葛解肌汤中葛根素的含量。方法:采用Hypersil BDS C_(18)色谱柱,以乙腈-0.1%磷酸水溶液为流动相梯度洗脱。流速为1.0 ml·min~(-1),检测波长250 nm,柱温30℃。结果:葛根素进样量在0.032 3~0.323 2μg范围内呈良好的线性关系(r=0.999 8,n=6);平均加样回收率为101.4%,RSD为2.9%(n=6)。结论:该法具有简便易行、检测灵敏、重复性好等优点,可用于柴葛解肌汤中葛根素的含量测定。
Objective: To establish a method for the determination of puerarin in Chaige Jieji Decoction by reversed-phase high performance liquid chromatography. Methods: Hypersil BDS C_ (18) column was used with mobile phase of acetonitrile-0.1% phosphoric acid as mobile phase. The flow rate was 1.0 ml · min -1, the detection wavelength was 250 nm and the column temperature was 30 ℃. Results: The injection volume of puerarin showed a good linearity (r = 0.999 8, n = 6) in the range of 0.032 3-0.3323 μg. The average recovery was 101.4% and the RSD was 2.9% (n = 6). Conclusion: The method is simple, sensitive, reproducible, and has the advantages of good reproducibility, which can be used for the determination of puerarin in Chaige Jieji Decoction.
