为了研究银杏叶多糖(PGBL)对肝纤维化的抑制作用,试验采用肿瘤坏死因子α(TNF-α)激活肝星形细胞之后,分别加入不同浓度(0.005,0.050和0.500 g/m L)的PGBL,用酶联免疫吸附测定(ELISA)法检测培养上清液中胶原蛋白Ⅲ水平。结果表明:0.050和0.500 g/m L的PGBL能明显抑制TNF-α刺激肝星形细胞的胶原蛋白Ⅲ的表达量,并且0.500 g/m L的PGBL可以保护TNF-α对肝星形细胞的刺激。说明PGBL对肝纤维化的修复作用可通过抑制TNF-α激活肝星形细胞和保护肝星形细胞来共同完成。
In order to study the inhibitory effect of Ginkgo biloba leaf polysaccharide (PGBL) on hepatic fibrosis, the hepatic stellate cells were activated with tumor necrosis factor alpha (TNF-α) and then treated with different concentrations of 0.005, 0.050 and 0.500 g / mL PGBL was detected by enzyme-linked immunosorbent assay (ELISA). The results showed that PGBL of 0.050 and 0.500 g / ml could significantly inhibit the expression of collagen III in hepatic stellate cells stimulated by TNF-α, and 0.500 g / ml of PGBL could protect TNF-α against hepatic stellate cells stimulate. These results suggest that the repair effect of PGBL on liver fibrosis can be achieved by inhibiting the activation of hepatic stellate cells and protecting hepatic stellate cells by TNF-α.
