MAO是一个分解体内生物活性单胺类的酶,存在于线粒体内。它在调节体内单胺类代谢中起着重要作用。研究表明,在许多病理情况下均明显改变MAO活力。我们借鉴国外经验,改建了一个敏感性高、特异性强的测定MAO的放射化学法。以[~3H]-5-羟色胺为底物与组织匀浆温育,产物以乙酸乙酯提取,直接在液体闪烁仪上测定。结果表明,酶浓度曲线的线性关系良好;酶的最适底物浓度为6×10~(-4)M;最适pH7.2~7.4;米氏常数(Km值)为2×10~(-4)M;用1.25毫克脑匀浆、2.5毫克肺匀浆即可满意测定MAO;用本方法测得大白鼠
MAO is an enzyme that breaks down the bioactive monoamines in the body and resides in the mitochondria. It plays an important role in regulating monoamine metabolism in vivo. Studies have shown that in many pathological conditions were significantly changed MAO activity. We learn from foreign experience, rebuilt a high sensitivity and specificity of the determination of MAO radiochemical method. To [~ 3H] -5-hydroxytryptamine as a substrate incubated with tissue homogenates, the product was extracted with ethyl acetate, measured directly on a liquid scintillometer. The results showed that the linear relationship of enzyme concentration curve was good. The optimal substrate concentration of enzyme was 6 × 10 -4 M, pH7.2-7.4, Km value was 2 × 10 ~ ( -4) M; with 1.25 mg brain homogenate, 2.5 mg of lung homogenate MAO can be measured satisfactorily; measured using this method, rats
