目的探讨血管内皮生长因子(VEGF)在血卟啉衍生物(HPD)加光动力作用(PDT)诱导人胰腺癌细胞PANC-1凋亡中的表达。方法应用流式细胞术检测细胞凋亡率,免疫组化染色法检测VEGF蛋白表达,RT-PCR检测基因表达水平。结果HPD+PDT实验组人胰腺癌细胞PANC-1凋亡率达(36.40±4.21)%,明显高于单纯HPD或PDT及空白三个对照组(P<0.05),VEGF蛋白和基因的表达均显著低于三个对照组(P<0.05),三个对照组间细胞凋亡、VEGF蛋白和基因表达率差异均无统计学意义。结论HPD+PDT可诱导人胰腺癌细胞PANC-1凋亡和抑制VEGF表达,进而抑制肿瘤血管生长。
Objective To investigate the expression of vascular endothelial growth factor (VEGF) in human pancreatic cancer cell PANC-1 induced by hematoporphyrin derivative (HPD) and photodynamic therapy (PDT). Methods The apoptosis rate was detected by flow cytometry. The expression of VEGF protein was detected by immunohistochemical staining. The gene expression level was detected by RT-PCR. Results The apoptosis rate of PANC-1 in HPD + PDT group was (36.40 ± 4.21)%, significantly higher than that in HPD + PDT group and blank control group (P <0.05). The expression of VEGF protein and gene in HPD + (P <0.05). There was no significant difference in apoptosis, VEGF protein and gene expression between the three control groups. Conclusion HPD + PDT can induce the apoptosis and inhibit the expression of VEGF in human pancreatic cancer cell line PANC-1, and then inhibit the tumor angiogenesis.
