目的:探讨巴戟天对卵巢切除所致大鼠骨质疏松症的治疗作用及其机理。方法:将60只雌性Wistar大鼠随机分为5组,即空白对照组、假手术组、模型组、阳性对照组和巴戟天组,每组12只。摘除大鼠双侧卵巢1个月后,灌胃给予巴戟天水煎剂,给药3个月。采用骨组织形态计量学的方法对大鼠胫骨不脱钙骨切片进行形态计量;采用免疫组化方法检测OB和MSC OPG、RANKL的蛋白表达。结果:与模型组比较,巴戟天组大鼠胫骨TBV%显著增高,TRS%以及TFS%、MAR明显降低,同时OB、MSCRANKL蛋白表达皆显著降低。结论:巴戟天对卵巢切除所致的大鼠骨质疏松症具有一定的治疗作用,能抑制OB和MSC RANKL的蛋白表达,是其能够治疗骨质疏松症的机理之一。
Objective: To investigate the therapeutic effect and mechanism of Morinda officinalis on osteoporosis induced by ovariectomy in rats. Methods: Sixty female Wistar rats were randomly divided into five groups: blank control group, sham operation group, model group, positive control group and Morinda officinalis group, 12 rats in each group. Rats were ovariectomized for 1 month and administered with Morinda officinalis decoction orally for 3 months. Morphometry of non-decalcified bovine tibia was performed by bone histomorphometry. The protein expressions of OPG and RANKL in OB and MSC were detected by immunohistochemistry. Results: Compared with the model group, the TBV% of the tibia in the Morinda officinalis group was significantly increased, the TRS% and TFS%, MAR were significantly decreased, while OB, MSCRANKL protein expression were significantly reduced. Conclusion: Morinda officinalis plays a role in the treatment of osteoporosis induced by ovariectomy in rats and inhibits the protein expression of RANKL in OB and MSC. It is one of the mechanisms that Morinda officinalis can treat osteoporosis.
