目的:探讨复方皂矾洗剂对雄激素性脱发的作用及机制。方法:60只雄性成年SD大鼠随机均分为空白组,模型组,米诺地尔酊0.05g/ml组,复方皂矾洗剂2g/ml组,复方皂矾洗剂0.5g/ml组,复方皂矾洗剂0.25g/ml组6组中,颈后皮下注射丙酸睾酮制造雄激素脱发大鼠模型。造模时予以背部外用药物,空白组、模型组均予以蒸馏水,余下4组分别予以0.05g/ml的米诺地尔酊,2g/ml、0.5g/ml、0.25g/ml的复方皂矾洗剂。计数皮肤横切面毛囊数量;测定皮肤中甘油三酯、游离脂肪酸、总胆固醇含量,评价不同浓度复方皂矾洗剂疗效。测定血液中睾酮、二氢睾酮的含量,测定皮肤中二氢睾酮的含量以及固醇调节元件结合蛋白(SREBP-1、SREBP-2)及含半胱氨酸的天冬氨酸蛋白水解酶-12(caspase-12)的表达,探讨复方皂矾洗剂作用机制。结果:复方皂矾洗剂2g/ml组中总毛囊和终毛毛囊数量较模型组增多,甘油三脂、游离脂肪酸、总胆固醇的含量较模型组降低,复方皂矾洗剂0.5g/ml组,复方皂矾洗剂0.25g/ml组作用程度依次减弱。2g/ml复方皂矾洗剂组皮肤中二氢睾酮的含量,SREBP-1、SREBP-2及caspase-12的表达均较模型组降低。结论:2g/ml、0.5g/ml、0.25g/ml的复方皂矾洗剂对雄激素性脱发均有治疗作用,以2g/ml为最佳剂量。复方皂矾洗剂的作用机制可能与降低皮肤中二氢睾酮的含量和SREBP-1,SREBP-2的表达,降低油脂分泌,抗脂质过氧化,抑制内质网应激,抑制毛囊内质网途径的凋亡有关。
Objective: To investigate the effect and mechanism of compound soap alum lotion on androgenetic alopecia. The content of dihydrotestosterone in the skin and the contents of steroid regulatory element binding protein (SREBP-1, SREBP-2) and cysteine-containing aspartate protease- 12 (caspase-12) expression, to explore the mechanism of compound soap alum lotion. Results: The total hair follicles and the final hair follicles in 2 g / ml compound soap lotion group increased more than the model group. The contents of triglyceride, free fatty acid and total cholesterol were lower than those in the model group. The compound soap group was 0.5 g / ml , Compound soap alum lotion 0.25g / ml group in turn decreased the degree of action. The content of dihydrotestosterone, the content of SREBP-1, SREBP-2 and caspase-12 in the skin of 2g / ml soap soap group were lower than those in model group. Conclusion: 2g / ml, 0.5g / ml, 0.25g / ml compound soap alum lotion have therapeutic effect on androgenetic alopecia, the best dose of 2g / ml. The action mechanism of compound soap alum lotion may reduce the content of dihydrotestosterone in the skin and the expression of SREBP-1, SREBP-2, reduce oil secretion, anti-lipid peroxidation, inhibit the endoplasmic reticulum stress and inhibit hair follicle endoplasmic reticulum Network of apoptosis related.
