应用比色法和电镜测定、观察了大鼠注射内毒素(L组)和氮芥+内毒素(NL组)后6个时相点血清血管紧张素Ⅰ转换酶(ACE)的活性变化及肺血管内皮细胞(EC)的形态改变。结果表明:L组于注射内毒素后1、5和12h血清ACE显著高于对照组(P<0.01),24h逐渐下降,至72h趋于正常;NL组于12、24hACE明显高于对照组(P<0.05)。L组各时相均见EC不同程度的肿胀、退变,重者坏死;NL组EC病变明显减轻。两组血清ACE的动态变化均与肺EC超微结构的病变程度密切相关。提示检测血清ACE活性变化对判断急性肺损伤、EC病变程度和了解ACE在血液循环调节方面的作用有着重要的意义。
Using colorimetric and electron microscopy, we observed the changes of serum ACE activity at 6 time points after injection of endotoxin (L group) and nitrogen mustard + endotoxin (NL group) Vascular endothelial cells (EC) morphological changes. The results showed that the serum ACE of L group was significantly higher than that of control group at 1, 5 and 12 hours after LPS injection (P <0.01), gradually decreased at 24 hours and then became normal at 72 hours after LPS injection. The levels of ACE at 12 and 24 hours in NL group were significantly higher than those in control group Group (P <0.05). In each group of L group, the swelling, degeneration and severe necrosis of EC were observed in varying degrees. The EC lesion in NL group was significantly reduced. The dynamic changes of serum ACE in both groups were closely related to the degree of lung EC ultrastructure. Prompt detection of changes in serum ACE activity to determine acute lung injury, EC lesions and to understand the role of ACE in the regulation of blood circulation is of great significance.
