目的建立血液中白藜芦醇的固相萃取/LC-MS/MS方法。方法自制固相萃取柱,在长约13cm的酸性滴定管中,底部填入约0.02g玻璃棉,依次加入柱填料PSA 0.15g,Florisil 0.3g,Silica 0.45g。对样本进行固相萃取,采用LC-MS/MS方法进行检测,运用保留时间和多反应监测(MRM)方式对血液中白藜芦醇进行定性定量分析。结果回收率为95.66%~98.58%,最低检测限(LOD)为1.79μg/mL,线性范围2.5~20μg/mL;相关系数为0.9997。结论本文所建方法,适用于血液中白藜芦醇的检测。
Objective To establish a method for the determination of resveratrol in blood by LC / MS / MS. Methods The homemade SPE column was filled with about 0.02 g glass wool in an acid burette of about 13 cm in length. The column filler PSA 0.15 g, Florisil 0.3 g and Silica 0.45 g were sequentially added. The samples were subjected to solid-phase extraction and detected by LC-MS / MS. Resveratrol in blood was qualitatively and quantitatively analyzed by retention time and multiple reaction monitoring (MRM). Results The recoveries ranged from 95.66% to 98.58%. The lowest limit of detection (LOD) was 1.79μg / mL and the linear range was 2.5 ~ 20μg / mL. The correlation coefficient was 0.9997. Conclusion The method proposed in this paper is suitable for the detection of resveratrol in blood.
