目的探讨实验性低血糖和血糖波动对糖尿病大鼠血清内皮素-1(ET-1)的影响。方法将正常Wistar大鼠随机分为对照(NC)组和低血糖(NL)组,糖尿病大鼠随机分为对照(DC)组、单纯低血糖(DL)组和低血糖后高血糖(DLH)组。用胰岛素诱导低血糖。DLH组于低血糖后给予50%葡萄糖灌胃诱发高血糖。所有实验大鼠于胰岛素或生理盐水注射前后1.5、4.5、6.5h测定血糖和ET-1。结果低血糖各组血清ET-1水平明显升高(DLH组>DL组>NL组);NL组ET-1水平在血糖最低点时最高(116.83±11.68)pg/ml;DL组和DLH组ET-1水平在血糖最低点时均无升高,峰值呈延迟现象;DLH组血糖再度升高时ET-1水平开始升高,峰值高于DL组[(239.3±22.22)pg/ml vs(161.12±55.94)pg/ml,P<0.05]。结论低血糖对糖尿病大鼠血管内皮损伤更明显;血糖剧烈波动对血管内皮损伤比持续低血糖更严重。
Objective To investigate the effects of experimental hypoglycemia and blood glucose fluctuation on serum endothelin-1 (ET-1) in diabetic rats. Methods Normal Wistar rats were randomly divided into control group (NC) and hypoglycemia group (NL). Diabetic rats were randomly divided into control group (DC), simple hypoglycemia group (DL) and hyperglycemia group group. Insulin-induced hypoglycemia. DLH group after hypoglycemia given 50% glucose gavage induced hyperglycemia. All experimental rats before and after insulin or saline injection 1.5,4.5,6.5 h determination of blood glucose and ET-1. Results Serum levels of ET-1 in hypoglycemia groups were significantly higher than those in DLH groups (DLH group> DL group> NL group). The level of ET-1 in NL group was the highest (116.83 ± 11.68) pg / ml at the lowest blood glucose level The ET-1 level did not increase at the lowest point of blood glucose, the peak value was delayed. In the DLH group, the level of ET-1 began to increase when the blood glucose was increased again, the peak value was higher than DL group [(239.3 ± 22.22) pg / ml vs 161.12 ± 55.94) pg / ml, P <0.05]. Conclusion Hypoglycemia is more obvious on vascular endothelial cells in diabetic rats. Severe fluctuations of blood glucose have more serious damage to vascular endothelial cells than persistent hypoglycemia.
