目的 在大鼠主动脉球囊损伤模型上 ,研究血管中三种一氧化氮合酶异构体 (神经型 ,内皮型和诱导型 )在内膜增殖过程中表达的变化。方法 2 0只Wistar大鼠分成 5组 ,损伤组 (n =16 ) :行胸腹主动脉球囊拉伤 ,术后 1、3、7、14天取材 ;对照组 (n =4) :不进行球囊损伤。应用蛋白印迹杂交的方法 ,观察主动脉损伤后内膜增殖过程中神经型 ,内皮型和诱导型一氧化氮合酶表达的变化。结果 在正常血管中 ,存在着神经型和内皮型一氧化氮合酶的表达 ;血管损伤后 1天 ,诱导型一氧化氮合酶出现表达 ,3天达高峰 ,较术后 1天增加 48% ,并持续到术后 14天。内皮型一氧化氮合酶在损伤后1、3、7、14天 ,表达较对照组分别增加 5 0 % ,10 5 % ,86 % ,49%。神经型一氧化氮合酶在内膜增殖过程中无动态改变。结论 血管损伤后 ,正常血管内皮细胞中内皮型一氧化氮合酶表达的上调 ,对抑制血管内膜增殖起一定的代偿作用 ;诱导型一氧化氮合酶表达的增加 ,在抑制内膜增殖方面起主要作用 ;神经型一氧化氮合酶与内膜增殖可能无关。
OBJECTIVE: To study the changes of expression of three nitric oxide synthase isoforms (neuronal, endothelium and inducible) in blood vessel during intimal hyperplasia in rat aortic balloon injury model. Methods Twenty Wistar rats were divided into five groups: injury group (n = 16): balloon-thoracoabdominal aortic balloon injury was performed on the 1st, 3rd, 7th and 14th days after operation. The control group (n = 4) Balloon injury. The expression of neuronal, endothelial and inducible nitric oxide synthase (NOS) in aortic intimal hyperplasia was observed by Western blotting. Results The expression of neuronal and endothelial nitric oxide synthase existed in normal blood vessels. The expression of inducible nitric oxide synthase occurred at 1 day after vascular injury and peaked at 3 days, which was 48% more than that at 1 day after operation , And continued to 14 days after surgery. Endothelial nitric oxide synthase increased by 50%, 105%, 86% and 49%, respectively, on the 1st, 3rd, 7th and 14th day after injury. Conclusion The up-regulation of endothelial nitric oxide synthase expression in normal vascular endothelial cells may play a compensatory role in inhibiting the proliferation of vascular intima after vascular injury.
