目的:研究1,25二羟维生素D3[1,25(OH)2D3]对Lscc02人喉鳞状细胞癌细胞系增殖及凋亡的影响。方法:将不同浓度的1,25(OH)2D3与Lscc02人喉鳞状细胞癌细胞系共同培养,采用MTT法、流式细胞仪及透射电镜观察该细胞系的增殖及凋亡诱导情况。结果:(10-10~10-7)mol/L浓度的1,25(OH)2D3均能抑制Lscc02细胞系的增殖,抑制作用随1,25(OH)2D3浓度的增加而增强,在半抑制浓度时,抑制作用随时间延长而增强。流式细胞仪显示,经1,25(OH)2D3作用的喉鳞状细胞癌细胞位于细胞周期G0/G1期的细胞数量明显增多且细胞凋亡数目明显增加。结论:1,25(OH)2D3体外能抑制Lscc02细胞系的增殖并诱导细胞凋亡,为临床探索1,25(OH)2D3治疗喉癌提供了重要的实验依据。
Objective: To investigate the effects of 1,25-dihydroxyvitamin D3 [1,25 (OH) 2D3] on the proliferation and apoptosis of Lscc02 human laryngeal squamous cell carcinoma cell line. Methods: Different concentrations of 1,25 (OH) 2D3 were co-cultured with Lscc02 human laryngeal squamous cell carcinoma cell line. The proliferation and apoptosis induction were observed by MTT, flow cytometry and transmission electron microscopy. RESULTS: 1,25 (OH) 2D3 (10-10 ~ 10-7) mol / L inhibited the proliferation of Lscc02 cells. The inhibitory effect was enhanced with the increase of 1,25 (OH) 2D3 concentration. Inhibition concentration increased with time when concentration was inhibited. Flow cytometry showed that the number of cells located in the G0 / G1 phase of the cell cycle increased significantly and the number of apoptotic cells increased significantly by 1,25 (OH) 2D3 laryngeal squamous cell carcinoma cells. CONCLUSION: 1,25 (OH) 2D3 can inhibit the proliferation and induce the apoptosis of Lscc02 cell line in vitro, which provides an important experimental basis for the clinical exploration of 1,25 (OH) 2D3 in the treatment of laryngeal cancer.
