目的:研究3’-大豆苷元磺酸钠对大鼠脑缺血再灌注损伤后视网膜作用的影响。方法:采用Longa方法制备大脑中动脉脑缺血再灌注损伤模型,一次性给予不同剂量3’-大豆苷元磺酸钠(1.0,2.0 mg.kg-1)舌下静脉注射。观察给药后脑梗死范围和脑组织含水量的影响。用夹闭大鼠一侧颈总动脉制备大鼠视网膜缺血模型。夹闭颈总动脉前,给药组分别舌下静脉注射不同剂量3’-大豆苷元磺酸钠(1.0,2.0 mg.kg-1)。观察对夹闭大鼠颈总动脉前及灌注后1 h眼底荧光血管造影和视网膜电流图的影响。结果:与模型组相比,1.0,2.0 mg.kg-1的3’-大豆苷元磺酸钠能显著降低脑组织含水量(P<0.05);与模型组比,1.0,2.0 mg.kg-1的3’-大豆苷元磺酸钠能使视网膜电流图振幅显著增加(3.72±1.12)mV,P<0.05;(4.87±1.37)mV,P<0.01,峰潜时显著减少(49.1±13.4)s,P<0.05;(33.6±12.5)s,P<0.01;与生理盐水组比,1.0,2.0 mg.kg-1的3’-大豆苷元磺酸钠能使眼底荧光血管造影视网膜动脉充盈时间显著减少(48.4±6.7)s,P<0.05;(35.7±3.4)s,P<0.01。结论:3’-大豆苷元磺酸钠对大鼠脑缺血再灌注损伤有明显的保护作用。3’-大豆苷元磺酸钠能扩张视网膜血管,对视网膜缺血再灌注损伤有明显的保护作用。
Objective: To study the effect of sodium 3’-daidzein sulfonate on the retinal function after cerebral ischemia-reperfusion injury in rats. Methods: The model of middle cerebral artery occlusion (MCAO) was established by Longa method. The rats were injected sublingual vein with different dosage of 3’-daidzein sodium sulfonate (1.0, 2.0 mg.kg-1) in one time. To observe the range of cerebral infarction and brain water content after administration. Rat retinal ischemia model was prepared by clipping the common carotid artery in rats. Before clamping the common carotid artery, different doses of 3’-daidzein sodium sulfonate (1.0, 2.0 mg.kg-1) were injected into the sublingual vein in the administration group. The effects of occlusion of the common carotid artery on rat retinal fluorescein angiography and retinal galvanogram 1 h after perfusion were observed. Results: Compared with the model group, 1.0, 2.0 mg.kg-1 sodium 3’-daidzein sulfonate significantly reduced brain water content (P <0.05), compared with the model group, 1.0, 2.0 mg.kg (3.72 ± 1.12) mV, P <0.05; (4.87 ± 1.37) mV, P <0.01, the peak latency was significantly decreased (49.1 ± 13.4) s, P <0.05; (33.6 ± 12.5) s, P <0.01; Compared with the saline group, 1.0,2.0 mg.kg-1 of 3’-daidzein sulfonate could make fundus fluorescein angiography retinal Arterial filling time was significantly reduced (48.4 ± 6.7) s, P <0.05; (35.7 ± 3.4) s, P <0.01. Conclusion: Sodium 3’-daidzein sulfonate has a significant protective effect on cerebral ischemia-reperfusion injury in rats. Sodium 3’-daidzein sulfonate can dilate the retinal blood vessels, which has a significant protective effect on retinal ischemia-reperfusion injury.
