白蒺藜有效组分对自发性高血压大鼠心肌纤维化的影响

目的观察白蒺藜有效组分对自发性高血压大鼠心肌纤维化的影响以探讨白蒺藜有效组分抑制高血压靶器官损伤的可能机制。方法32只14周龄雄性自发性高血压大鼠随机分为模型组、卡托普利组、白蒺藜组、联用组,每组8只,同周龄雄性Wistar大鼠8只作为正常对照。卡托普利组采用卡托普利每只30mg·kg-.1d-1、白蒺藜组采用白蒺藜有效组分每只15mg·kg-.1d-1、联用组采用白蒺藜有效组分每只15mg·kg-1.d-1和卡托普利每只30mg·kg-1.d-1、模型组及正常组给0.9%NaCl溶液每只2ml/d灌胃,共8周。采用尾动脉容积法测定血压;分离左心室,计算左心室重量指数;生化方法检测各组大鼠心肌羟脯氨酸含量。结果与正常Wistar大鼠比较,14周龄SHR大鼠收缩压明显升高(P<0.01)。8周后,模型组大鼠收缩压较14周龄时明显升高(P<0.05);与模型组相比,白蒺藜组收缩压明显下降(P<0.01),左心室重量指数则无明显差异(P>0.05);卡托普利组、联用组收缩压显著下降(P<0.01),左室重量指数显著降低(P<0.05);各用药组心肌羟脯氨酸含量均明显降低(P<0.05),心肌胶原沉积减少。结论白蒺藜有效组分能降低SHR收缩压,改善左室肥厚程度,降低心肌胶原含量,从而保护高血压病所致的靶器官损伤。 Objective To observe the effect of effective components of Radix Paeoniae Alba on myocardial fibrosis in spontaneously hypertensive rats to explore the possible mechanism of effective components of Radix Paeoniae Alba on the inhibition of target organ damage in hypertension. Methods Thirty-two 14-week-old male spontaneously hypertensive rats were randomly divided into model group, captopril group, baiyao group and combination group, 8 in each group. Eight male Wistar rats of the same age were used as normal controls. . Captopril group used 30 mg·kg-.1d-1 captopril, 15 mg·kg-.1d-1 group each of effective components of Radix Paeoniae Alba group and effective components of Radix Paeoniae Alba Each 15 mg·kg-1.d-1 and captopril 30 mg·kg-1.d-1 each, the model group and the normal group were given intragastric administration of 0.9% NaCl solution 2 ml/d for 8 weeks. The caudal artery volumetric method was used to measure the blood pressure; the left ventricle was isolated and the left ventricular mass index was calculated; the myocardial hydroxyproline content was detected by biochemical methods. RESULTS Compared with normal Wistar rats, systolic pressure was significantly increased in 14-week-old SHR rats (P<0.01). After 8 weeks, the systolic blood pressure of the model group was significantly higher than that of the 14-week-old group (P<0.05). Compared with the model group, the systolic blood pressure of the white peony group significantly decreased (P<0.01), while the left ventricular weight index was not significant. Differences (P>0.05); Significantly decreased systolic blood pressure (P<0.01) and left ventricular mass index (P<0.05) in captopril group and combination group; Myocardial hydroxyproline content was significantly decreased in each drug group (P<0.05), myocardial collagen deposition decreased. Conclusion The effective components of Radix Paeoniae Alba can reduce the SHR systolic blood pressure, improve the left ventricular hypertrophy and reduce the content of collagen in the heart muscle, so as to protect the target organ damage caused by hypertension.
