目的 探讨反义转化生长因子(TGF) β1基因转染对骨肉瘤细胞MG 63表达基质金属蛋白酶(MMPs)和尿激酶型纤溶酶原激活因子(uPA )等肿瘤转移相关因子的影响。方法 将反义TGF β1基因转染MG 63 ,G418筛选转染细胞后,采用Northernblot法检测转染细胞MMP 2、MMP 9和uPA的表达情况。结果 反义TGF β1基因转染骨肉瘤细胞上清培养的Mv 1 Lu细胞增殖活性为0 .975±0 .0 16(对照组1.0 3 2±0 .0 2 7,P >0 .0 5 ) ;Northernblot法检测显示MMP 2和uPAmRNA的表达明显降低。结论 通过反义基因技术阻断骨肉瘤细胞TGF β1自分泌环,在降低肿瘤细胞TGF β1表达的同时,也降低了转移相关因子MMP 2、uPA的表达,从而可能抑制肿瘤的侵袭和转移
Objective To investigate the effect of antisense transforming growth factor (TGF) β1 gene transfection on the expression of matrix metalloproteinases (MMPs) and urokinase-type plasminogen activator (uPA) in osteosarcoma MG63 cells. Methods Antisense TGF β1 gene was transfected into MG63 and G418 cells. The expression of MMP 2, MMP 9 and uPA in transfected cells was detected by Northern blot. Results The proliferation activity of Mv 1 Lu cells transfected with antisense TGFβ1 gene in osteosarcoma cell supernatant was 0.975 ± 0.016 (1.032 ± 0.227, P> 0.05) ; Northern blot analysis showed that the expression of MMP 2 and uPA mRNA was significantly reduced. Conclusion Blocking the autocrine loop of TGFβ1 in osteosarcoma cells by antisense gene technology can reduce the expression of TGFβ1 and decrease the expression of metastasis-related factors MMP 2 and uPA, which may inhibit tumor invasion and metastasis
