目的探讨mir-99a在上皮性卵巢癌(EOC)患者中的表达与临床病理特征及预后的关系。方法采用实时荧光定量PCR(qRT-PCR)法检测137例EOC患者组织中mir-99a的表达水平,分析mir-99a表达与卵巢癌患者的临床病理资料的相关性。采用Kaplan-Meier法进行生存分析并绘制生存曲线,生存因素采用log-rank检验。结果 EOC患者组织中mir-99a的表达较癌旁正常组织降低(P<0.001)。mir-99a的低表达与高组织学分级(P=0.027)、淋巴结转移(P=0.001)、腹膜转移(P<0.001)和较高的FIGO分期(P<0.001)有关。Kaplan-Meier分析与log-rank检验结果显示,mir-99a的低表达与EOC患者生存期短有关(对数秩检验,P=0.005)。多因素分析结果显示,mir-99a表达是患者总体生存的独立预测因子(HR为2.368,95%CI:1.621,10.925,P=0.016)。结论mir-99a的表达是上皮性卵巢癌患者独立的预后因素,mir-99a可能成为临床实践中一项新的诊断和预后标志物。
Objective To investigate the relationship between the expression of mir-99a in epithelial ovarian cancer (EOC) patients and clinicopathological features and prognosis. Methods Real-time quantitative PCR (qRT-PCR) was used to detect the expression of mir-99a in 137 patients with EOC. The correlation between the expression of mir-99a and the clinicopathological data of ovarian cancer patients was analyzed. Survival analysis and survival curves were performed by Kaplan-Meier method. The survival factors were analyzed by log-rank test. Results The expression of mir-99a in EOC patients was lower than that in normal tissues (P <0.001). The low expression of mir-99a was associated with high histological grade (P = 0.027), lymph node metastasis (P = 0.001), peritoneal metastasis (P <0.001) and higher FIGO stage (P <0.001). Kaplan-Meier analysis and log-rank test showed that the low expression of mir-99a correlated with the short survival of patients with EOC (log-rank test, P = 0.005). Multivariate analysis showed that mir-99a expression was an independent predictor of overall patient survival (HR 2.368, 95% CI: 1.621, 10.925, P = 0.016). Conclusion The expression of mir-99a is an independent prognostic factor in patients with epithelial ovarian cancer. Mir-99a may be a new diagnostic and prognostic marker in clinical practice.
