目的 建立肝癌细胞凋亡模型 ,探讨顺铂抗癌作用机理 ,为进一步研究细胞凋亡分子机制打下基础。方法 用抗癌药顺铂诱导培养的人肝癌细胞株SMMC - 772 1肝癌细胞发生凋亡 ,用四氮甲唑蓝比色试验 (MTT) ,细胞形态学检查 ,DNA凝胶电泳检测。结果 顺铂可显著抑制SMMC - 772 1细胞的生长 ,并有很好的量效和时效关系 ,经药物作用后 ,肝癌细胞在显微镜下呈现典型的凋亡形态学改变 ,DNA琼脂糖凝胶电泳呈现明显的梯度带。结论 用抗癌药顺铂能成功诱导人肝癌细胞株SMMC - 772 1肝癌细胞发生凋亡 ,该细胞凋亡模型的建立 ,将有助于进一步探讨肝癌细胞凋亡的分子机制
Objective To establish apoptotic model of hepatocellular carcinoma and explore the anticancer mechanism of cisplatin, which lays the foundation for further study of molecular mechanism of apoptosis. Methods Apoptosis of hepatocellular carcinoma cell line SMMC-7721 induced by anticancer drug cisplatin was detected by MTT assay, cytomorphology and DNA gel electrophoresis. RESULTS: Cisplatin significantly inhibited the growth of SMMC-7721 cells, and had a good dose-effect and time-dependent relationship. After the drug was applied, the liver cancer cells showed typical apoptotic morphological changes under the microscope. DNA agarose gel electrophoresis Shows a clear gradient band. Conclusion The anticancer drug cisplatin can successfully induce the apoptosis of human hepatocellular carcinoma cell line SMMC-7721. The establishment of this apoptosis model will help further explore the molecular mechanism of hepatocellular carcinoma cell apoptosis.
