目的本研究通过分析比较遗传性癫痫大鼠(Tremor,TRM)与正常大鼠(Wistar)海马中BDNF蛋白表达与定位差异,进一步阐明癫痫发病机制。方法以正常Wistar大鼠为对照组,应用PCR技术,筛选阳性自发性癫痫大鼠TRM作为实验组;通过Western blot技术分别对TRM及Wistar大鼠海马中BDNF蛋白进行半定量检测;免疫组织化学技术分别测定TRM及Wistar大鼠海马中BDNF蛋白的定位表达变化。结果与Wistar大鼠相比,TRM大鼠的海马中BDNF蛋白表达无明显变化;BDNF蛋白在海马DG区和CA1区定位表达减少。结论与Wistar大鼠相比,TRM大鼠海马DG区和CA1区BDNF蛋白定位表达减少,表明癫痫的发作可能与BDNF蛋白在海马区定位的异常变化有关。
OBJECTIVE: In this study, the differences of BDNF protein expression and localization in the hippocampus between the hereditary epilepsy rats (Tremor, TRM) and normal rats (Wistar) were compared to further clarify the pathogenesis of epilepsy. Methods Normal Wistar rats were used as control group. PCR was used to screen the TRM of positive spontaneous epilepsy rats as experimental group. Western blot was used to detect the expression of BDNF protein in hippocampus of TRM and Wistar rats respectively. Immunohistochemistry The changes of the localization of BDNF protein in hippocampus of TRM and Wistar rats were measured respectively. Results Compared with Wistar rats, the expression of BDNF protein in hippocampus of TRM rats did not change significantly. The expression of BDNF protein in hippocampus DG and CA1 area decreased. Conclusion Compared with Wistar rats, the expression of BDNF protein in DG and CA1 areas of hippocampus in TRM rats decreased, indicating that the onset of epilepsy may be related to the abnormal changes of BDNF protein in hippocampus.
