目的:探讨哮喘小鼠气道和肺组织中半乳糖凝集素8(Galectin-8)及基质金属蛋白酶9(MMP-9)的表达情况及布地奈德对其表达的影响。方法:36只清洁级雌性BALB/c小鼠随机分为对照组、哮喘组、布地奈德干预组,建立卵清蛋白(OVA)致敏、激发的哮喘模型,观察各组小鼠肺组织的病理变化;免疫组化、RT-PCR分别测定各组小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达;结果 :哮喘组小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达显著高于对照组及布地奈德干预组(P<0.05),Galectin-8蛋白和MMP-9蛋白的表达呈正相关(r=0.89,P<0.05)。结论:哮喘小鼠气道和肺组织中Galectin-8、MMP-9蛋白和mRNA的表达明显升高,并且两者蛋白的表达具有相关性;布地奈德干预后Galectin-8、MMP-9蛋白和mRNA表达下调。
OBJECTIVE: To investigate the expression of Galectin-8 and MMP-9 in airway and lung tissues of asthmatic mice and the effect of budesonide on its expression. Methods: Thirty-six female BALB / c mice of clean grade were randomly divided into control group, asthma group and budesonide intervention group. Ovalbumin (OVA) sensitized and challenged asthmatic model was established. The expression of Galectin-8, MMP-9 protein and mRNA were detected by immunohistochemistry and RT-PCR in airway and lung tissue of mice in each group.Results: Galectin-8 , While the expression of MMP-9 protein and mRNA was significantly higher than that of the control group and budesonide intervention group (P <0.05). The expression of Galectin-8 protein and MMP-9 protein were positively correlated (r = 0.89, P <0.05). Conclusion: The expression of Galectin-8, MMP-9 protein and mRNA in asthmatic mice airway and lung tissue were significantly increased, and the expression of Galectin-8, MMP-9 protein And down-regulated mRNA expression.
