目的探讨白介素-10(interleukin-10,IL-10)对转化生长因子-β(transtorm growth factor beta,TGF-β)刺激成纤维细胞增殖、胶原分泌以及向肌成纤维细胞(myofibroblast,MFB)转化的影响。方法培养人胚肺成纤维细胞,根据不同的处理方法分为五组:A组加入TGF-β(5 ng/ml),B组加TGF-β(5 ng/ml)+IL-10(5 ng/ml),C组加TGF-β(5 ng/ml)+IL-10(15 ng/ml),D组加TGF-β(5 ng/ml)+IL-10(25 ng/ml),E组为空白对照组(无细胞)。于72 h使用MTT方法观察各组成纤维细胞增殖情况,羟脯氨酸消化法检测胶原产生量,免疫细胞化学法检测平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)的表达情况。结果B、C、D组细胞内α-SMA表达量分别与A组比较,差异均有统计学意义(P均<0.01),D组与B、C组比较差异均有统计学意义(P均<0.01);羟脯氨酸产生量C与B组、D组比较差异均有统计学意义(P均<0.01)。细胞增殖吸光度D组与C、B组比较差异有统计学意义(P<0.01)。结论在体外IL-10对TGF-β刺激成纤维细胞的增殖、胶原产生及α-SMA表达有抑制作用,且呈剂量依赖性。
Objective To investigate the effects of interleukin-10 (IL-10) on proliferation, collagen secretion and myofibroblast (MFB) fibroblast proliferation induced by transforming growth factor-β (TGF-β) Impact. Methods Human embryonic lung fibroblasts were cultured and divided into five groups according to different treatments: group A received TGF-β (5 ng / ml), group B received TGF-β (5 ng / ml) + IL-10 (5 ng / ml) and IL-10 (15 ng / ml) in group C and 25 ng / ml TGF-β (5 ng / ml) , E group as blank control group (no cell). The proliferation of fibroblasts was observed by MTT method at 72 h. The collagen production was detected by hydroxyproline digestion method. The expression of α-smooth muscle actin (α-SMA) was detected by immunocytochemistry. Results The expression of α-SMA in group B, C and D was significantly different from that in group A (P <0.01), and there was significant difference between group D and group B and C (P <0.01). The amount of hydroxyproline C was significantly different from those of group B and group D (all P <0.01). Cell proliferation absorbance D group and C, B group difference was statistically significant (P <0.01). Conclusion IL-10 inhibits the proliferation, collagen production and α-SMA expression of fibroblast stimulated by TGF-β in a dose-dependent manner.
