优化了苯酚-硫酸法测定仙人掌多糖含量的条件:取多糖样品2 m1,加4%苯酚0.5 m1,迅速加入浓硫酸5 ml,摇匀后静置5 min,于482 nm处测定吸光度。多糖在7.5~37.5μg/ml浓度范围内与吸光度的线性关系良好。采用果胶酶和纤维素酶双酶提取法从仙人掌中提取仙人掌多糖,最佳提取条件为:果胶酶浓度0.7%,纤维素酶浓度0.3%,40℃提取1.5 h。仙人掌多糖产率为20.08%,纯度为83.02%。
The optimum conditions for the determination of polysaccharides content of cactus by phenol-sulfuric acid method were as follows: 2 ml of polysaccharide sample, 0.5 ml of 4% phenol and 5 ml of concentrated sulfuric acid were added rapidly, and the mixture was allowed to stand for 5 min after shaking. The absorbance was measured at 482 nm. The linear relationship between polysaccharide and absorbance in the range of 7.5 ~ 37.5μg / ml was good. The extraction of cactus polysaccharides from cactus by pectinase and cellulase double enzyme extraction was the best extraction conditions: pectinase concentration of 0.7%, cellulase concentration of 0.3%, 40 ℃ for 1.5 h. Cactus polysaccharide yield of 20.08%, a purity of 83.02%.
