目的采用HPLC-ELSD测定桑树不同各部位中的1-脱氧野尻霉素(DNJ)含量,并测定四川省4个代表性蚕桑基地的桑树各部位DNJ的含量。方法色谱柱为DIKMA Ineresil NH2column(250 mm×4.6 mm,5μm);流动相为乙腈-甲醇-水(70:10:20);流速1ml.min-1,柱温35℃。结果线性范围1.02~12.24μg(r=0.9995);平均回收率为98.7%(RSD=1.58%)。结论不同种不同产地桑树中DNJ含量差异较大,绵阳三台县和泸州泸县的山桑干皮含量最高(分别为8.145、7.624 mg.g-1),桑叶含量居中,桑椹含量为最低(0.345 mg.g-1)。
Objective To determine the content of 1-deoxynojirimycin (DNJ) in different parts of mulberry by HPLC-ELSD and determine the content of DNJ in four parts of mulberry in Sichuan province. Methods The column was DIKMA Ineresil NH2column (250 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-methanol-water (70:10:20). The flow rate was 1 ml.min-1 and the column temperature was 35 ℃. Results The linear range was 1.02 ~ 12.24μg (r = 0.9995). The average recovery was 98.7% (RSD = 1.58%). Conclusion The contents of DNJ in mulberry trees of different habitats are quite different. The contents of mulberry leaves in Santai County and Luzhou County in Mianyang City are the highest (8.145 and 7.624 mg.g-1, respectively), the content of mulberry leaves is the highest and the content of mulberry is the lowest (0.345 mg.g-1).
